In vitro study | The circulation of Tenofovir is actively secreted by the kidneys through glomerular filtration and renal tubules. Tenofovir is not a substrate for human organic cation transport type 1 (hOCT1) or hoct2. In MRP4 overexpressing cells, Tenofovir accumulates to five-fold lower levels, and MRP inhibitors can increase its accumulation. Tenofovir does not significantly affect mitochondrial DNA(mtDNA) in human hepatoblastoma (HCC) cells, skeletal muscle cells (SkMCs) or renal proximal tubule epithelial cells. In HepG2 cells or SkMCs, Tenofovir increased lactate production by less than 20%. Tenofovir is a potent phosphorylation of Tenofovir diphosphate (TFV-DP) in HepG2 cells and human primary hepatocytes. In the cell-based assay, the 50% effective concentration of Tenofovir anti-HBV was 1.1 mM, with a 50-fold increase in efficiency after the addition of the pre-bis-isoproxil group. In Vitro and clinically, Tenofovir has full activity against lamivudine-resistant HBV. Tenofovir inhibited the proliferation of hepatogenic hepatocellular carcinoma cells and normal skeletal muscle cells with CC(50) values of 398 μm and 870 μm, respectively. Tenofovir has a weaker effect than cidofovir on the proliferation and viability of renal proximal tubular epithelial cells, and cidofovir has a potential related nucleotide analog to induce renal tubular dysfunction. |
